Not sure I understand the question entirely.  You need to replace 
libusb0.dll with the new one in the .zip file.  Leave the rest 
of  the libusb0 driver (.sys, .inf, etc) alone -- so do not uninstall it etc.

I do this via:

copy libusb0.dll libusb0.dll.old
unzip libuisb-ddk.zip
